Figure 1. (a) Restriction map of pHS41. The restriction sites and the number of each site indicate the approximate position in the vector. The arrow indicates the orientation of each DNA fragments assembled. LB: left border of T-DNA; RB7MAR: matrix attachment region of tobacco; UbiPro: maize ubiquitin promoter; bar: gene for phosphinothricin acetyltransferase; Nos: terminator of nos (nopaline synthase) gene; MSP1: oil palm mesocarp-specific promoter; TP: oil palm transit peptide of Δ9-stearoyl-ACP-desaturase; RB: right border of T-DNA.
(b) Restriction endonuclease analysis of pHS41. The predicted fragments (bp in length) for each endonuclease digestion are listed as follows: 2 fragments (8405, 1594) for SpeI, 1 fragment (9999) for AscI, 5 fragments (2495, 2423, 1960, 1570, 1551) for BglII and 2 fragments (7372, 2627) for NheI. The observed fragments in each digestion were of the same size as predicted.
