This strategy involves restriction digestion ofgenomic DNA from mutant plant with an appropriate enzyme (preferably with an enzyme which does not cut within the T-DNA). Self ligated molecules are used for transformation of E. coli. The self-ligated fragments containing the T-DNA along with the flanking sequences are able to survive and are rescued as plasmid because of the presence of origin of replication (Ori), shown here in blue.
